چکیده

(S)-Hydroxynitrile lyase from Hevea brasiliensis (HbHnl) (EC 4.1.2.39) catalyzes the reversible synthesis of chiral cyanohydrins from aldehydes or ketones and HCN. The enzymatic formation of (S)-mandelonitrile (MN) from benzaldehyde (BA) and HCN was studied in two-phase systems of buffer and organic solvents (diisopropyl ether, methyl-t-butyl ether) using a Lewis cell to investigate the interaction between mass transfer and the biocatalytic reaction. The enzymatic reaction rate in the aqueous phase saturated with organic solvents is drastically reduced in comparison to pure buffer due to the increase of the Michaelis-Menten constants of the substrates. Mass transfer of the substrates from the organic to the aqueous phase and mass transfer of the product in the opposite direction could be described by the two-film theory. The formation of (S)-mandelonitrile in the Lewis cell follows an aqueous phase distributed reaction model, which means that the enzymatic reaction takes place in the bulk of the aqueous phase and in the thin film close to the interface and/or directly at the interface. Using the Hatta number it could be shown that the mass transfer of benzaldehyde from the organic to the aqueous phase is enhanced by the biocatalytic reaction of the (S)-hydroxynitrile lyase from Hevea brasiliensis.

Purification and Characterization of Alginate Lyase from Mucoid Pseudomonas aeruginosa Strain 214

Pseudomonas aeruginosa is an opportunistic pathogen that causes a variety of infections in compromised patients. The ability of Pseudomonas aeruginosa to produce chronic infection is based in part on its ability to biosynthesis of biofilm, and alginate is the major polysaccharide in the synthesized biofilm. So alginate degradation is very essential in the dispersion of Pseudomonas aeruginosa bi...

Biochemical and computational study of an alginate lyase produced by Pseudomonas aeruginosa strain S21

Objective(s): Alginates play a key role in mucoid Pseudomonas aeruginosa colonization, biofilm formation, and driving out of cationic antibiotics. P. aeruginosa alginate lyase (AlgL) is a periplasmic enzyme that is necessary for alginate synthesis and secretion. It also has a role in depolymerization of alginates. Using AlgLs in cystic fibrosis patients along with anti...

One-step purification and characterization of alginate lyase from a clinical Pseudomonas aeruginosa with destructive activity on bacterial biofilm

Objective(s): Pseudomonas aeruginosais a Gram-negative and aerobic rod bacterium that displays mucoid and non-mucoid phenotype. Mucoid strains secrete alginate, which is the main agent of biofilms in chronic P. aeruginosa infections, show high resistance to antibiotics; consequently, the biological disruption of mucoid P. aeruginosa biofilms is an attractive area of study for researchers. Algin...

The Effect of Kinetics Parameters on Gold Extraction by Lewis Cell: Comparison between Synthetic and Leach Solution

Modern hydrometallurgical techniques have been adopted to produce high-purity gold. Many extractants were used for the extraction of gold from various synthetic solutions from which DiButyl Carbitol (DBC) had a unique superiority. Kinetics of process was studied in this paper and the influence of several parameters such as stirring speed, agitation time, impeller type, interfacial area, [Au...

Comparative expression of wild-type and highly soluble mutant His103Leu of hydroxynitrile lyase from Manihot esculenta in prokaryotic and eukaryotic expression systems.

Low protein solubility and inclusion body formation represent big challenges in production of recombinant proteins in Escherichia coli. We have recently reported functional expression of hydroxynitrile lyase from Manihot esculenta, MeHNL, in E. coli with high in vivo solubility and activity using directed evolution. As a part of attempts to clarify the mechanism of this phenomenon, we have desc...
